I just watched Ju-On: The Curse for a second time. I've seen the other three Japanese Ju-Ons, but I've forgotten a lot of the story.
For anyone who knows this film and the other Japanese Ju-Ons extremely well, I'm curious if you have a theory for any of the following (and maybe the other films explain some of this and I just forgot):
BEWARE OF MASSIVE SPOILERS IF YOU HAVEN'T WATCHED THIS FILM YET!

It seems clear (although maybe I'm wrong) that the Saeki's and the Murakami's lived in the same house. Did the Murakami's live there before the Saeki's or after? It seems like they must have lived in the house after the Saeki's, otherwise, how could Toshio appear in his ghostly form to Tsuyoshi's girlfriend in the classroom and how could Kayako kill Yuki? However, if they lived in the house after the Murakami's, why wouldn't the real estate agent be aware of this? That seems inexplicable.

Who does the real-estate agent's sister see at the end of the film, and what is the significance? It's an awfully abrupt ending. This one is probably answered in Ju-On 2: The Curse 2 (as I remember #2 beginning by replaying a large chunk of the ending of this one), but I can't remember the story. If it's plainly answered there, then maybe don't tell me, because it might be a spoiler for when I rewatch #2.

I guess we can suppose that Kanna's mom would have died or gone crazy or something, but any theories on what happened to Tsuyoshi?
Even with these things unexplained, I like this film a lot, and actually raised my rating a point (to a 9) on this viewing. A few of the scenes are incredibly well done, such as Mizuho's ordeal in the school, Kanna's appearance back home in her ghostly state and the extended sequence ending with Kobayashi's last appearance, including Takeo's phone call and disturbing "baggage".

I can answer question number three:
Tsuyoshi disappeared without a trace. As julian1190 said, his fate is covered in a short film by Shimizu entitled "4444444444"
In the newspaper clippings that the real extate agent Tatsuya's sister (Kyoko, the one with the psychic powers) got from her co-worker in the park, it was described how Noriko (Tsuyoshi & Kanna's mother; Yuki's aunt) was found inside the entrance to her home with her internal organs ripped out Boy, am I glad they didn't SHOW that! LOL!

Yes, they lived in the house after the Saeki's. Remember they kept getting mail for Takeo, the previous owner.

Who does the real-estate agent's sister see at the end of the film, and what is the significance? It's an awfully abrupt ending. She sees Kayako, in general. Maybe she knows
spoilers
Kayako took over another person.
end spoilers

I guess we can suppose that Kanna's mom would have died or gone crazy or something, but any theories on what happened to Tsuyoshi?
This is sorta made clear in the short film 44444444, which is available on The Grudge Unrated DVD. In it, Tsuyoshi also finds the evil cell phone and meets the same end as Mizuho.
BTW, in the Ju-on video side Manga, Kanna and Noriko suffer extremely gruesome fates
