That was a short film Rodriguez shot and sent to Frank Miller in order to persuade him to allow Rodriguez to direct a film adaptation of his stories.

The Hitman is a reoccurring character in the Sin City books. I believe the opening sequence was just to introduce him, so when you see him at the end of the movie in the elevator with Becky, you know she's going to end up dead real soon.
