1. How did the Real Estate Agent girl know that Sake determines whether or not there's spirits nearby?
2. Did Kayako get reincarnated and buy the house??? I'm confused.

Sorry, I haven't been logging onto IMDb recently LOL!

1. The lady who determines ghosts with the Sake was not the woman from the real estate office, it was the sister of the real estate agent, and she was apparently "gifted" in that manner This is gone into greater detail in "Ju-On 2."
2. This question (I assume you mean the creepy looking woman peering from behind the curtain) is answered in "Ju-On 2"

As has been stated by the above poster, the sister can sense presence of spirits.
She said herself to her brother that, in ancient times, Sake were used to determinate if there where spirits in a certain place.
