The part where Pee-wee is dressed as a nun and his eyes bug out when Kevin Morton says, "I want the orphanage to have my bike."
When Pee-wee is hitchhiking and throws the big rubber thumb into the traffic out of frustration.
Pee-wee is stumbling around in the dark after being booted out of Mickey's car, he hears coyotes barking and howling and almost laughs as he says, "Kitty?" -

Lol !. I'm cracking up thinking about the part where Pee Wee keeps knocking on the door at the Buxtons house and the music that plays as that happens !!. And the conversation with the butler is hilarious . It looks like the butler wants to laugh during that part too !.

I always thought it was funny how Pee-Wee's movie about him is nothing like his adventure, but he doesn't seem to mind at all.
Or, the dramatic camera move when Francis goes, "You'll be sorry, Pee-Wee Herman!"

Love this movieI love when him and Simone are talking about their dreams and she asks him "Do you have any dreams, Pee Wee?" and his response is priceless "I'm rolling a big doughnut and there's this giant snake". I love that line. Also when Pee Wee and Mickey get past the checkpoint dressed in their disguises and when they drive down the highwayMickey is giving Pee Wee (dressed as a woman (sort of)) the slight lust look (he has been in prison)hillarious

- The Mr. T cereal he nibbles, ignoring the rest of that huge breakfast
- The stereotypical BMX kids
- Pee-Wee the Nun- "I'll say, I'm going to start a paper route right now!"
- "I took a knife and I- (sees Pee-Wee's face) You know those 'do not remove under the penalty of law' labels they put on mattresses? Well I cut one off!"
- That huge chain he wrapped around the bike several times.
- "Oh really? Where they hosing him down!?"

"Too late! Chip!" (crooked grin) "Uh, it looks like a pen."
Pee Wee nearly losing him mind before jumping in the pool ("Go ahead and scream your head off, we're miles from where anyone can hear you!") Followed by him trying to escape through a locked window.
How exited he got at the magic store after hearing there was a box for Pee-Wee. I'm 46 and if you tell me I got a box in the mail - I'll do the same thing.
Micky talking to himself during the drive, ending with "Heh!"

That whole chase scene makes me laugh.
I think it's the sight of Santa and Godzilla in the sleigh and Godzilla's arm is around Santa's shoulder.

I just watched this on netflix today for the first time in I don't know how many years, and I couldn't help but laugh at some of the innuendo in this movie that completely went over my head when I saw this as a kid. One example is when Pee Wee and Simone are talking in the dinosaur and PW tells her to follow her dream of moving to Paris and not letting anything stop her. Her response, with her jealous boyfriend listening from outside: "No one's ever put it to me that way." That line and the look on the bf's face made me crack up.
