It's already been remade.in 1993, directed by Christopher Guest with Daryl Hannah as the 50 Foot Woman. I doubt you'll be getting another remake.

I don't know about the main character, but I think Tracy Lords would be pretty good evil girlfriend of the husband character.

Are we talking a halfway serious effort or pull-out-all-the-stops campy?
Kristen Bell, perhaps. Playing an introverted little woman with hidden anger issues. Make the radiation capable of turning people, not necessarily taller, but into whatever their id wants them to be (large and fearsome, in her case).
And Dax Shepard as her rotter of a husband.
Maybe Sophia Bush as the girlfriend not nearly as evil as clueless and manipulated.
